Mass Effect 3 multiplayer can impact single-player
As of now, we know the multiplayer portion is a standalone mode and supports up to four players. You'll can join up to three other players in an elite Special Forces squad, sent to liberate "key territories from enemy control." A neat feature is being able to choose your class, race, weapons, and special abilities, integrating some of that personal customization feel into the multiplayer, even though you won't be playing as Shepard. The co-op missions are playable over Xbox LIVE, PSN, and the Internet (for PC), though it doesn't sound like local co-op will be supported.
Some of you may also be interested to know that the Mass Effect 3 multiplayer is actually developed by BioWare's new Montreal studio, rather than the core team.
Another question Priestly answered is whether the multiplayer will affect single-player. The short answer to that is "yes," but it's a bit more complex than that. Mass Effect 3 has this new Galaxy at War system, which measures your "Galactic Readiness" level based on how well-prepared Shepard is to face the Reapers. How you utilize your resources -- people, weapons, fleets, and so forth -- directly affects your rating. Now, maximum Galactic Readiness can be achieved in the campaign, but players can actually use multiplayer as an alternate method of reaching optimal readiness.
